This family is universal in eukaryotes, and best known to phosphorylate nucleoside diphosphates to nucleoside triphosphates. They have a ferridoxin-like fold, and use a phospho-histidine intermediate during catalysis (the donor phosphate is transiently attached to a His residue on the NDPK). There are 9 NDPK homologs in human, of which four (NME1-4) are orthologous to Drosophila awd. NME2 (NDPK-B) was shown to phosphorylate the KCa3.1 potassium channel calcium channel on a conserved histidine [1] and a G protein β subunit [2]. Several other mammalian proteins are reported to be histidine phosphorylated, including histone H4, Gβ1, annexin-1, P-selectin, and ornithine transcarbamylase, some of which may be substrates for NDPKs (no other human histidine kinases are known).
Serine kinase activity was also reported for NME2 [3], though the transfer appeared to be stoichiometric (i.e. no turnover: one enzyme, one phosphorylation action) and so termed a phosphotransferase. The Drosophila homolog, awd, was also seen to autophosphorylate and transphosphorylate (ovalbumin) on Serine [4] and an E. coli NDPK was also shown to autophosphorylate on both serine and histidine [5].
